Texture Analysis applied to second harmonic generation image data for diseases classification and Multi-View Second Harmonic Generation Imaging

Remodeling of the extracellular matrix has been implicated in many diseases. To quantitate the remodeling, we implement a form of texture analysis to delineate the collagen fibrillar morphology observed in second harmonic generation microscopy images for ovarian cancer and idiopathic pulmonary fibrosis classification. To explore the morphology of extra-cellular matrix in 3D and extend our capability for 3D texture analysis, we utilize reflective micro-prisms to enable multi-view SHG imaging by redirecting the focused excitation and collection of subsequent emission. Improved imaging of 3D collagen structure is demonstrated possible with multi-view SHG microscopy. Further we developed 3D texture analysis adapting for multi-view second harmonic generation imaging platform.
